The list below was compiled by searching the NCI Clinical Trials data base with the term “breast – Male” in the “anatomic_sites” field using the published API, and restricting the results to Active trials with a status change in the last 6 months, which is hopefully the best way currently to find the newest trials (improvements coming in the database for this, I understand from the NCI’s API webpage.

This list therefore may contain the most recent trials to come on line for which men with breast cancer are eligible. Please explore the trials, and read details on their main pages on the NCI webpage, via the links provided.

Durvalumab and Combination Chemotherapy in Treating Patients with Stage I-III Triple Negative Breast Cancer

Single Arm Neoadjuvant Phase I / II Study of MEDI4736 (Anti-PD-L1 Antibody) Concomitant with Weekly Nab-Paclitaxel and Dose Dense Doxorubicin / Cyclophosphamide (AC) Chemotherapy for Clinical Stage I-III Triple Negative Breast Cancer

Phase I_II

This phase I / II trial studies the side effects and best dose of durvalumab when given together with paclitaxel albumin-stabilized nanoparticle formulation, doxorubicin hydrochloride, and cyclophosphamide and to see how well they work in treating patients with stage I-III breast cancer that does not have estrogen receptors, progesterone receptors, or large amounts of human epidermal growth factor receptor (HER)2 / neu protein. Monoclonal antibodies, such as durvalumab, may interfere with the ability of tumor cells to grow and spread. Drugs used in chemotherapy, such as paclitaxel albumin-stabilized nanoparticle formulation, doxorubicin hydrochloride, and cyclophosphamide, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving durvalumab together with paclitaxel albumin-stabilized nanoparticle formulation, doxorubicin hydrochloride, and cyclophosphamide may kill more tumor cells.

A Study Evaluating the Safety and Efficacy of Venetoclax in Combination With Trastuzumab Emtansine in Patients With Previously Treated HER2-Positive Locally Advanced or Metastatic Breast Cancer

A Phase lb / ll, Randomized, Double-Blind, Placebo-Controlled, Multicenter Study Evaluating the Safety, Tolerability, Pharmacokinetics, and Efficacy of Venetoclax in Combination With Trastuzumab Emtansine in Patients With Previously Treated HER2-Positive Locally Advanced or Metastatic Breast Cancer

Phase I_II

This two-part study is composed of two stages: a Phase Ib stage consisting of a dose-escalation phase and an expansion phase; and a Phase II, randomized, placebo-controlled, double-blind, multicenter stage. The Phase Ib stage will assess the safety and tolerability, determine the maximum tolerated dose (MTD) and the recommended Phase II dose (RP2D), and evaluate the preliminary efficacy of trastuzumab emtansine in combination with venetoclax in participants with previously treated human epidermal growth factor receptor 2 (HER2) positive unresectable locally advanced breast cancer (LABC) or metastatic breast cancer (MBC). Additional patients may be enrolled in an expansion phase to evaluate the safety, tolerability, and efficacy of trastuzumab emtansine in combination with venetoclax at RP2D in patients with previously treated HER2-positive LABC or MBC who have previously received either trastuzumab emtansine or trastuzumab deruxtecan (DS-8201a). The Phase II randomized stage will evaluate the safety, efficacy, tolerability, and pharmacokinetics of trastuzumab emtansine in combination with venetoclax at RP2D compared with trastuzumab emtansine plus placebo in participants with previously treated HER2-positive LABC or MBC who have not received prior trastuzumab emtansine therapy, either alone or in combination with other anti-cancer therapies.

FES-PET / CT Scans for the Optimization of Tamoxifen Dose in Patients with Estrogen Receptor Positive, ESR1 Mutant Metastatic or Unresectable Breast Cancer

A Pilot Study of FES Imaging to Optimize Tamoxifen Dose for Metastatic Breast Cancer Patients with ESR1 Mutations

Phase O

This early phase I trial studies how well FES-PET / CT scans work in determining the best dose of tamoxifen for treating patients with estrogen receptor positive, ESR1 gene mutant breast cancer that has spread to other places in the body (metastatic) or cannot be removed by surgery (unresectable). FES is a low-dose radioactive form of estrogen and can “light up” where estrogen receptor positive cancer is present in the body. Diagnostic procedures such as FES-PET / CT, may help to determine the optimal dose of tamoxifen and prevent treatment resistance due to underdosing on standard dose of tamoxifen in patients with breast cancer.

Study of Alpelisib (BYL719) in Combination With Trastuzumab and Pertuzumab as Maintenance Therapy in Patients With HER2-positive Advanced Breast Cancer With a PIK3CA Mutation

EPIK-B2: A Two Part, Phase III, Multicenter, Randomized (1:1), Double-blind, Placebo-controlled Study to Assess the Efficacy and Safety of Alpelisib (BYL719) in Combination With Trastuzumab and Pertuzumab as Maintenance Therapy in Patients With HER2-positive Advanced Breast Cancer With a PIK3CA Mutation

Phase III

The purpose of this two parts multicenter, randomized, double-blind, placebo-controlled, Phase III study is to evaluate the efficacy and safety of alpelisib compared to alpelisib matching-placebo in combination with trastuzumab and pertuzumab as maintenance treatment of patients with HER2-positive advanced breast cancer whose tumor harbors a PIK3CA mutation following induction therapy with a taxane in combination with trastuzumab and pertuzumab. Part 1 is the open-label, safety run-in part of the study, designed to confirm the recommended phase 3 dose (RP3D) dose of alpelisib in combination with trastuzumab and pertuzumab. Following Part 1, Part 2 will be initiated, which is the randomized, Phase III part of the study.

ASTX727 and Talazoparib for the Treatment of Triple Negative or Hormone Resistant / HER2-Negative Metastatic Breast Cancer

A Phase I Study of ASTX727 plus Talazoparib in Patients with Triple Negative or Hormone Resistant / HER2-Negative Metastatic Breast Cancer

Phase I

This phase I trial studies the side effects and best dose of ASTX727 and talazoparib in treating patients with triple negative or hormone resistant / HER2-negative breast cancer that has spread to other places in the body (metastatic). ASTX727 is a combination of two chemotherapy drugs, cedazuridine and decitabine, and may increase the immune system’s response to cancer and talazoparib. Talazoparib is an anticancer drug called a PARP (poly ADP ribose polymerase) inhibitor and is in development for the treatment of a variety of cancers. The combination of ASTX727 and talazoparib may help boost the immune system to stop or reverse the growth of tumors.

Pembrolizumab and Binimetinib in Treating Patients with Locally Advanced or Metastatic Triple Negative Breast Cancer

Phase I / II Trial of Pembrolizumab in Combination with Binimetinib in Unresectable Locally Advanced or Metastatic Triple Negative Breast Cancer

Phase I_II

This phase I / II trial studies the best dose of pembrolizumab and binimetinib and how well it works when given together with pembrolizumab in treating patients with triple negative breast cancer that has spread to other parts of the body (metastatic). Immunotherapy with monoclonal antibodies, such as pembrolizumab, may help the body’s immune system attack the cancer, and may interfere with the ability of tumor cells to grow and spread. Binimetinib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Giving pembrolizumab and binimetinib may work better in treating patients with triple negative breast cancer.

Galunisertib and Paclitaxel in Treating Patients with Metastatic Androgen Receptor Negative or Triple Negative Breast Cancer

A Phase Ib Trial of LY2157299 (TGFβR1 Kinase Inhibitor) with Paclitaxel in Patients with Triple Negative Metastatic Breast Cancer

Phase I

This phase I trial studies the side effects and best dose of galunisertib when given together with paclitaxel in treating patients with androgen receptor negative or triple negative breast cancer that has spread to other places in the body. Some tumors need growth factors, which are made by the body’s white blood cells, to keep growing. Galunisertib may interfere with growth factors and help cause tumor cells to die. Drugs used in chemotherapy, such as paclitaxel, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving glunisertib together with paclitaxel may kill more tumor cells.

A Phase Ib Trial of Fulvestrant, Palbociclib (CDK4 / 6 Inhibitor) and Erdafitinib (JNJ-42756493, Pan-FGFR Tyrosine Kinase Inhibitor) in ER+ / HER2- / FGFR-Amplified Metastatic Breast Cancer (MBC)

Phase I

This phase Ib trial studies the side effects and best dose of erdafitinib when given together with fulvestrant and palbociclib in treating patients with estrogen receptor positive, HER2 negative, and FGFR amplified stage IV breast cancer that has come back or cannot be removed by surgery. Drugs used in chemotherapy, such as fulvestrant, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Palbociclib and erdafitinib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Giving fulvestrant, palbociclib, and erdafitinib may work better in treating patients with breast cancer.

Lymph Node Dissection and Radiation Therapy in Treating Patients with Breast Cancer Previously Treated with Chemotherapy and Surgery

A Randomized Phase III Trial Comparing Axillary Lymph Node Dissection to Axillary Radiation in Breast Cancer Patients (cT1-3 N1) Who Have Positive Sentinel Lymph Node Disease after Neoadjuvant Chemotherapy

Phase III

This randomized phase III trial studies lymph node dissection and radiation therapy to see how well it works compared to radiation therapy alone in treating patients with breast cancer previously treated with chemotherapy and surgery. Lymph node dissection may remove cancer cells that have spread to nearby lymph nodes in patients with breast cancer. Radiation therapy uses high-energy x rays or protons to kill tumor cells. It is not yet known if radiation therapy works better alone or with lymph node dissection in treating patients with breast cancer previously treated with chemotherapy and surgery.

A Phase II Trial of Atezolizumab (Anti-PDL1) with Carboplatin in Patients with Metastatic Triple Negative Breast Cancer

Phase II

This randomized phase II trial studies how well carboplatin with or without atezolizumab works in treating patients with stage IV triple negative breast cancer. Drugs used in chemotherapy, such as carboplatin, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Immunotherapy with monoclonal antibodies, such as atezolizumab, may help the body’s immune system attack the cancer, and may interfere with the ability of tumor cells to grow and spread. Giving carboplatin with atezolizumab may work better in treating patients with stage IV triple negative breast cancer.

This phase I trial studies the side effects and best dose of alpha-TEA when given together with trastuzumab and to see how well they work for the treatment of HER2+ breast cancer that does not respond to treatment (refractory) and has spread to other places in the body (metastatic). Anti-cancer treatment, such as alpha-TEA, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Alpha-TEA may also alter cancer growth by stimulating the body’s immune response against the tumor. Trastuzumab is a form of “targeted therapy” because it works by attaching itself to specific molecules (receptors) on the surface of cancer cells, known as HER2 receptors. When trastuzumab attaches to HER2 receptors, the signals that tell the cells to grow are blocked and the cancer cell may be marked for destruction by the body’s immune system. Giving alpha-TEA and trastuzumab may work better for the treatment of HER2+ refractory and metastatic breast cancer compared to usual treatment.

A Phase 2 Study of Copanlisib (BAY 80-6946) in Combination with Fulvestrant in Patients with Metastatic Breast Cancer Progressing after Aromatase Inhibitor plus CDK 4 / 6 Inhibitor

Phase I_II

This phase I / II trial studies the side effects and how well copanlisib works when given together with fulvestrant in treating patients with estrogen receptor positive (ER+) and human epidermal growth factor receptor 2 negative (HER2-) breast cancer that has spread to other places in the body (advanced) and progressing after prior treatment. HER2 and ER are two types of proteins called receptors that can affect the growth of breast cancer cells. Additionally, investigators hope to learn from this study if tumor genetic information is important for predicting whether this type of breast cancer will respond to fulvestrant and copanlisib. Copanlisib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Hormone therapy using fulvestrant may fight breast cancer by blocking the use of estrogen by the tumor cells. Giving copanlisib and fulvestrant may work better in treating patients with ER+ and HER2- breast cancer compared to fulvestrant alone.

This trial studies how well paclitaxel, trastuzumab, and pertuzumab work in eliminating further chemotherapy after surgery in patients with HER2-positive stage II-IIIa breast cancer who have no cancer remaining at surgery (either in the breast or underarm lymph nodes) after pre-operative chemotherapy and HER2-targeted therapy. Drugs used in chemotherapy, such as paclitaxel, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Trastuzumab is a form of “targeted therapy” because it works by attaching itself to specific molecules (receptors) on the surface of tumor cells, known as HER2 receptors. A Phase 2 Study of ONC201 in Recurrent / Refractory Metastatic Breast Cancer and Advanced Endometrial Carcinoma

Phase II

Background: The new drug ONC201 have been shown to kill breast cancer and endometrial cancer cells in the laboratory. The exact mechanism of action is not completely clear yet, but the ONC201 destroys the mitochondria inside the cells. Blocking mitochondrial activity may kill tumor cells, which would shrink tumors. Researchers want to see if ONC201 helps shrink tumors of certain breast or endometrial cancers and if that effect is maintained. Objective: To see if ONC201 shrinks tumors with a lasting effect. Eligibility: Adults ages 18 and older who have metastatic breast cancer (hormone-positive or triple-negative) or metastatic endometrial cancers. Design: Participants will be screened with: Medical history Physical exam Heart, blood, and urine tests CT and bone scans Review of medical report and tumor sample Participants will have a tumor biopsy before starting treatment and after 5 weeks taking the study drug. A scan or ultrasound may be used to guide the biopsy. Patients will receive local anesthetic and a needle will remove a small piece of tumor. The study will be done in 28-day cycles. Every day 1 of each cycle participants will repeat most screening tests, will be seen by the physician and receive a supply of the study drug. Participants will take the study drug by mouth once every 7 days. They will keep a diary of when they take the drug and any side effects. During cycle 1, participants will get weekly calls to discuss their health and symptoms. Images will be repeated every 2 cycles to evaluate reponse to the treatment. …
